dc.descriptionBacillus thuringiensis (Bt) is a Gram-positive bacterium characterized by the production of proteins with insecticidal activity, like Cry proteins in the form of crystalline parasporal bodies during its sporulation phase and other proteins in the vegetative phase of its growth. Bt and its toxins have been used in agriculture as a biological alternative against plague insects. The main characteristics of these toxins are their high specificity and innocuousness for vertebrates and other beneficial insects. While it is true that there are studies that evaluate some conditions for the growth of the microorganism and the production of its proteins, the results obtained have shown a discrepancy of data regarding the production of proteins and have not been evaluated at the same time operating and culture conditions to obtain maximum yield. Hence the importance of establishing an optimized protocol to produce large amounts of protein toxins from Bacillus thuringiensis var. kurstaki. In this work, two experimental Plackett Burman designs were carried out to evaluate the influential variables in the production of Bt proteins. In the first Plackett Burman, 7 influential variables were obtained, and in the second design, developed under the same conditions, of the 7 variables, CoCl2 and FeSO4 concentrations and temperature were statistically influential. Three Compound Central Designs were made (rotational DCC 2k) which generated three response surfaces showing the optimal values for protein production. The mathematical model generated by the response surfaces allowed to deduce that at a concentration of 30,83 mg L of CoCl2, 1,37 mg L of FeSO4 and a temperature of 34,9 degree centigrade, the total protein production calculated was maximum and took a value of 4,0 mg mL. Experimentally, using these culture and operating conditions, protein concentrations between 3,6 and 4,5 mg mL were obtained, which shows that both, mathematical model and the values obtained experimentally, are an approximate representation of the influence of CoCl2, FeSO4 and the temperature versus the production of total proteins from Bacillusen
