dc.descriptionThe cell wall of plants is made up of several polysaccharides, including cellulose, hemicellulose, and pectin. The cellulose, in turn, possesses crystalline and amorphous regions, which are hydrolyzed by the action of enzymes named as cellulases, endoglucanases and exoglucanases. The bacterium Pectobacterim carotovorum is notorious for its maceration to the cell wall of plants causing soft rot. The ability to destroy plants is due to the expression and secretion of a wide range of hydrolytic enzymes due to the action of the induction system of Gram-negative bacteria, which includes cellulases and expansins. P. carotovorum PC1 FJ890936 was used for the design of primers for amplification of the cellulase gene and to include the cell pelB export site. The PCR product was cloned into the expression vector pET22b-PcExl1 and transformed into Escherichia coli. The molecular weight of each of the clones obtained was 1267 bp. estimating the molecular weights of the expressed protein was performed using SDS-PAGE analysis and two bands of 39 kDa and 32 kDa, respectively. The enzymatic activity was carried out by a reducing sugar release reduction assay. However, cellulase did not show a possible interaction with the expansin PcExl1.
