dc.descriptionCancer is considered one of the pathologies with the highest mortality and morbidity rates in the world. The diagnosis is made in later stages, thus increasing the mortality rate. Melanoma is the most aggressive and deadly type of skin cancer, with 132 000 new cases diagnosed and 66 000 deaths annually. On the other hand, research in recent years has shown that RNA binding proteins (RBPs) play an important role in the modulation of specific genes that affect the initiation, development and progression of various types of cancer. Although, the molecular mechanism through which these proteins act is not known clearly, the results from ex vivo and xenographic models experiments suggest that the RBP UNR induces initiation and metastasis in melanoma. The objective of this work was to evaluate the UNR expression in melanoma samples from Ecuadorian individuals and associate this with demographic, histological and target genes that appear to be regulated by UNR. Thirty samples of paraffin embedded tumors were collected from patients with melanoma and twenty nine with nevus from ‘’Eugenio Espejo Hospital’’. Immunohistochemistry was applied using the Anti-CSDE1 antibody (ab96124) as well as qPCR to determine the level of UNR and 5 target genes that can be regulated by this RBP 5: PTEN, TGFBI, SERPINE1, S100A10 and GUSβ expression. A higher expression of UNR was obtained in tumors than in nevus, although a relationship between the protein and gene expression of UNR was not found, possibly due to a translation dependent on previously described IRES sequences in this gene.
