Honey from the Meliponini Tribe plays a fundamental role in medicinal use due to its high pharmacological potential and nutritional value. However, studies in the country about its biological properties and chemical composition are limited. For this reason, in the present study it was established to determine the antimicrobial capacity and the presence of the defensin peptide in stingless honey from the main meliponicultor provinces of the country. The antimicrobial activity of 35 honeys from the provinces of Loja, Pastaza, Tungurahua and El Oro was analyzed, to verify the significant differences of antimicrobial activity of all the honeys were compared in ANOVA for multiple comparisons between samples. On the other hand, honeys treated with proteinase k showed different antimicrobial effect. Furthermore, the total proteins were quantified by Bradford, the protein profiles of the honeys were analyzed in SDS-PAGE polyacrylamide gels and the presence of the defensin peptide was analyzed by Western Blot. The protein profiles showed more abundant band sizes, mainly between 100-50 kDa of proteins related to the plant and the stingless bee. The low molecular weight protein fractions are related to the most characterized and studied antimicrobial peptides in Apis mellifera, particularly the defensin-1 peptide. Finally, the presence of this 5.5 kDa peptide was determined in five honeys from stingless bees from the provinces of El Oro (OR15, OR18, OR20, OR25) and Pastaza (PS17). Therefore, it is concluded that meliponid honeys from the provinces of Ecuador showed promising results of antimicrobial activity and protein fractions.en
