dc.description | Ecuador is located in the Pacific Ring of Fire. For this reason, it is a country characterized by a large diversity of geothermal sources of volcanic origin or geological faults. Actually they are use as therapeutic and recreational, due to its physico-chemical components, which have been studied over time. However, there is no greater information about the microbial biodiversity of the geothermal sources in the country. For this reason the objective of this study was the isolation and identification of thermophilic bacteria from the geothermal source Papallacta located in Napo province. The temperature and in-situ pH of the terma was approximately 53°C and 7.1, respectively. The samples were obtained from water and sediment from the eye of the terma, for identification from DNA total, under the massive sequencing. Subsequently, within the process of bacterial isolation, samples were processed, inoculated and purified in semi-specific culture media. The bacterial isolates were characterized through amplification of 16S rRNA gene and sequencing. A total of 22 bacterial strains with similar morphology were isolated and high production of spores. The results of the massive sequencing-Illumina indicated a greater number of OTUs in the Proteobacteria phylum in both water and in sediment with a percentage of 47.5% and 69.7% respectively. Inside the edge Proteobacteria the most predominant class was Alpha-proteobacteria (50 genders) and that are known to be mostly culturable and proliferating bacteria favorably in aquatic environments. The bacteria identified in the study are directly related to the diversity of ecosystems in Ecuador and the world. The magnitude of existing bacteria in the Papallacta geothermal source encourages to future studies to explore other extreme environments and their microbial variety of biotechnological interest. | en |
