Resumen : En el presente trabajo de titulación se logró establecer material vegetal in vitro de semillas y brotes, partiendo de mortiño Vaccinium floribundum Kunth…
Descripción : In the present titling work, it was possible to establish in vitro vegetative material from seeds and shoots of mortiño (Vaccinium floribundum Kunth). For the in vitro establishment of seeds, different variables that influence in germination were tested, such as growth medium, photoperiod and temperature. The best treatments were T3 (MS + 24 hours light + 18 degrees Celsius) and T7 (WPM + 24 hours light + 18 degrees Celsius), which indicates that light hours, and temperature are the most influential factor in germination. Another important factor was the growth medium, the seedlings cultivated in WPM medium presents a higher vigor than the others plants. For the desinfection of seeds a reported protocol in 2010 by Torres et al. Was used, the following procedure were applied it starts with a desinfection in 70 percent for three minutes, then the material were rinsed with Sterile distilled water, later it was desinfected with 2.5 percent sodium hypochlorite plus Tween 20 for 10 minutes, finally it rinsed with sterile distilled water for five times. For this study the previously disinfected seeds were planted in Petri dishes (20 per box) with different growth mediums. For the seedlings different disinfection protocols were tested, the best treatment was T2 (With predisinfection + 70 percent alcohol for 1 minute + 2 percent NAClO for 20 minutes), the disinfected seedlings were seeded in sample tubes with WPM, to determine the best conditions for seedlings different variables such as contamination, oxidation and number of viable shoots were analyzed. Finally for leaves no results were obtained due to the age of the explant, since for this test leaves of adult plants were used this leaves underwent oxidation and died, also there was not a callus formation. For the statistical analysis the data were evaluated with a randomized block desing with factorial arrangement and LSD Fisher significance tests.
