dc.description | Spirulina sp. is a filamentous cyanobacteria with high nutritional value, its empirical use as dietary supplement has been used since the antiquity. The popularity of this food is due to its high protein content. It is present in various ecosystems including alkaline lakes. The present investigation was carried out in the Laguna de Colta intervened antropically, considered a high Andean lagoon of Ecuador located at 3420 m.s.n.m. The purpose of this work was the identification, isolation and purification of native strains of Spirulina spp. sampling methodology was performed by zones and random sample collection. Identification was done through a collection of dichotomous keys. The isolation was done by microscopy, dilutions, liquid - solid media and reisolated. Purification was performed with an increase of sodium bicarbonate, combination of dilutions with microscopic isolation and addition of cycloheximide. The growth conditions were 2500-3000 lux of artificial light, a photoperiod of 12h at a temperature of 21 4 Grades Celsius and continuous aeration. The results obtained from sampling indicated that of the 6 designated areas, Zone 1 with 36 samples was the area with greater Spirulina presence. Taxonomically, Spirulina subsalsa and Spirulina major were identified through morphological and microscopic characteristics. Thus it was determined that these native strains are adapted to anthropic interventions typical of this lentic ecosystem. The application of simple purification strategies allowed to establish monocultures of a genus, with the presence of the two native species. Which could be used commercially, after studies of their nutritional characteristics | en |
