dc.descriptionThe pollination of native and cultivated plants is achieved by the interaction of bees, nevertheless these insects can be affected by fungi and bacteria. This type of infection is dispersed in the colony by spores. Bacterial diseases such as American and European foulbrood are caused by Paenibacillus larvae and Melissococcus plutonius respectively, and the fungus disease Ascospherosis is caused by Ascosphaera apis. These diseases have high prevalence during bees' larval stage. Microbiological and parasitological are limited techniques for identification and diagnosis of bacterial and fungus bees’ diseases due to the transport, collection and incorrect processing of samples. As an alternative, molecular biology techniques such as PCR has better results, because is more effective in time, sensitivity and specificity. Consequently, a molecular method based on multiple PCR was used to identify these microorganisms. A protocol implemented with DNAzol was used for the extraction of DNA from larvae. For the gene amplification of the 3 microorganisms with bands of 973 bp, 281 bp and 136 bp Specific primers were used. The results of the multiplex PCR standardization showed high specificity of the primers (100 percent) and a detection limit of 0.01 pg of the PCR, except for A. apis with 0.01 fg. The obtained results shown the need the need to have a molecular method for identification of pathogens that cause diseases that affect beekeeping in Ecuador.en
