Resumen : Pectobacterium carotovorum es una bacteria patógena de plantas, que coloniza cultivos y vegetales con alto impacto agrícola y causa la podredumbre blanda.Mediante el mecanismo de autoinducción esta bacteria secreta proteínas como las expansinas, que pueden brindar una ventaja durante la infección a vegetales. Sin embargo, aunque la principal función de las expansinas es la de modificar la pared celular vegetal, se desconoce el rol que cumple la expansina PcExl1 en la infección a vegetales y el sitio específico de unión en el tejido vegetal. En este estudio, con el propósito de determinar el sitio especifico de unión de PcExl1, se expresó heterologamente PcExl1 y se realizaron experimentos de unión de la expansina PcExl1, marcada con el fluoróforo Cy5, con los xilemas aislados de apio, kale, verdolaga, betabel, ruibarbo y acelga. En los ensayos de microscopía se identificaron dos tipos de fibras: unas fibras helicoidales y otras fibras lineales y se observó que la expansina se une a las estructuras helicoidales que conforman los haces vasculares del xilema de los vegetales. Al estudiar la preferencia de unión de PcExl1 por un determinado tipo de fibra, se determinó que las fibras helicoidales aunque presentan una baja diferencia de cristalinidad del 5 por ciento con respecto a las fibras lineales, presentan diferencias del 29,1 FWHM (FWHM, del inglés full width at half maximum) en cuanto a la textura. Además, durante los experimentos de composición química de azucares de las fibras, se determinaron diferencias del 3 por ciento de xilosa y del 2 por ciento de arabinosa-galactosa entre las fibras helicoidales y las fibras lineales. Estas diferencias en la textura, así como en la relación de azúcares determinan la unión preferencial de la expansina a las fibras helicoidales. Descripción : Pectobacterium carotovorum is a pathogenic bacterium of plants, which colonizes crops and vegetables with high agricultural impact and causes soft rotting. By the mechanism of quorum sensing this bacteria secretes proteins, like the expansins, that can give an advantage during the infection to vegetables. Although, the main function of the expansins is to modify the plant cell wall, the role of the expansin PcExl1 in the infection to plants and the specific site of binding in plant tissue is unknown. In this study, for the purpose of determining the specific site of PcExl1 binding, PcExl1 was heterologously expressed and binding experiments of the PcExl1 expansin, labeled with the fluorophore CY5, were performed with the isolated xylems of celery, kale, purslane, beet, rhubarb and chard. In the microscopy tests two types of fibers were identified: helical fibers and other linear fibers and it was observed that the expansin is united to the helicoidal structures that conform the vascular bundles of the xylem of the vegetables. When studying the preference of PcExl1 binding to a certain type of fiber, it was determined that the helical fibers, although they have a low crystallinity difference of 5 percent with respect to linear fibers, show differences of 29.1 FWHM (FWHM, of english full width at half maximum) in terms of texture. In addition, during the chemical composition experiments of fiber sugars, differences of 3 percent of xylose and 2 percent of arabinose-galactose were determined between helical fibers and linear fibers. Differences in texture as well as in the ratio of sugars determine the preferential binding of the expansin to helical fibers.
