Descripción : Vasconcellea is the least studied genus of Caricaceae, it‟s commercial and biotechnological potential is underestimated and studies in this respect must be undertaken. Sixteen of the 21 species of this genus are distributed in Ecuador. The objective of the present work was to establish a protocol of induction of somatic embryogenesis in “highland papaya” Vasconcellea pubescens. The protocol involves two stages, the induction of somatic embryogenesis and the maturation of embryogenic calli. Two trials were performed with induction medium (IND) at different concentrations of 2,4-D (Trial 1: 4.5, 10, 67.5 y 112. mg/L and Trial 2: 0.5, 2.5, 4.5, 10 mg/L) in order to induce somatic embryogenesis on zygotic embryos from Vasconcellea pubescens. Further, were used different compositions of medium for development of embryos considering two maturation medium (MAT 1 without hormone and MAT 2 with 0,019 mM BAP and 0,023 mM NAA) and the induction medium from which the embryogenic calli were originally obtained. In all the number of somatic embryos and the weight of embryogenic calli were determined. A RCBD was used, the results were analyzed with R-Project using the proper statistical model. The zygotic embryos showed response in IND between 5 to 7 days after seeding on the medium. During the induction (Trial 1) the highest calli weight was obtained at 4.5 mg/L of 2,4-D while for the Trial 2 was obtained at 2.5 mg/L. During maturation (Trial 1) the highest embryogenic calli weight and the higher number of embryos were obtained from calli that originated from medium IND 4.5 mg/L of 2,4-D both from medium MAT 1, while for the Trial 2 the highest weight and number of embryos were with calli that originated from medium IND 10.0 mg/L of 2,4-D. In conclusion, the range of 2,4-D concentrations suitable for the induction of embryogenesis and further generation of somatic embryos of Vasconcellea pubescens was between 2.5 to 10 mg/L while higher concentrations elicit an inhibitory response. This behavior is similar to that seen in papaya.
