dc.descriptionSulfonamides (SAs) are an extensive group of synthetic antibiotics, used as veterinary drugs in cattle for the treatment and prevention of diseases. Residues of these antibiotics are secreted into milk that reaches the consumer; they cause harmful effects on human health that is why the identification and quantification of these compounds in milk are a priority. The aim of this work was to develop and implement a method for the identification and quantification of sulfamethazine and sulfamerazine in raw milk by Ultra Performance Liquid Chromatography (UPLC), because Ecuador had not developed a quantitative method for the identification of these two antibiotics in milk. The first method in this project was to standardize the chromatographic reading method, by using standards of sulfamethazine and sulfamerazine with 99% purity, and then it proceeded to perform calibration curves of each of the (SAs), by the triplication analysis of each standard. Then, the samples of raw milk were fortified with known concentrations of the two antibiotics, the extraction and purification of the samples was performed by solid phase extraction (SPE). By the method standardization the samples of raw milk were analyzed from the Alóag Parish in the Mejía Canton at the Pichincha Province. Retention time of each sulfonamide was obtained, a correlation coefficient close at 1 was achieved so that the linearity of the method was demonstrated, also a statistical study was performed to determine the limits of detection and quantification of the two ( SAs). After these limits were obtained they showed that the developed method is able to detect concentrations of parts per billion level. An average recovery rate of 91.05% and 115.71% for sulfamethazine and sulfamerazine, these were acceptable values for this indicator. Three of the 6 milk samples were positive for sulfamethazine, with values that exceed the LRM allowed by the Codex Alimentarius.en
dc.description.abstractLas sulfonamidas (SAs) son un extenso grupo de antibióticos sintéticos, utilizados como medicamentos veterinarios en el ganado vacuno para el tratamiento y prevención de enfermedades. Los residuos de este tipo de antibióticos son segregados a la leche que llega al consumidor causando efectos nocivos en la salud humana, por este motivo se vuelve una prioridad la identificación y cuantificación de estos compuestos en la leche. El objetivo de este trabajo fue desarrollar e implementar un método para la identificación y cuantificación de sulfametazina y sulfamerazina en leche cruda por medio de cromatografía liquida de ultra alta resolución (UPLC), ya que en el Ecuador no se había desarrollado un método cuantitativo para la identificación de estos dos antibióticos en la leche. En este proyecto primero se estandarizó el método de lectura cromatográfica mediante el uso de estándares, tanto de sulfametazina como sulfamerazina al 99% de pureza y se procedió a realizar curvas de calibración de cada una de las (SAs), esto mediante el análisis por triplicado de cada estándar. Seguidamente se fortificaron muestras de leche cruda con concentraciones conocidas de los dos antibióticos, la extracción y purificación de las muestras se realizó mediante extracción en fase sólida (SPE). Estandarizado el método, se corrieron muestras de leche cruda provenientes de la Parroquia Alóag del Cantón Mejía Provincia de Pichincha. Se obtuvo los tiempos de retención de cada sulfonamida, se consiguió un coeficiente de correlación cercano a 1 con lo que se demostró la linealidad del método, también se realizó un estudio estadístico con el que se determinó los límites de detección y cuantificación de las dos (SAs), obtenidos estos límites se demostró que el método desarrollado puede detectar concentraciones a nivel de partes por billón. Se obtuvo un porcentaje promedio de recuperación de 91.05% para sulfametazina y 115.71% para sulfamerazina, que son valores aceptables para este indicador. De las 6 muestras de leche analizadas 3 resultaron positivas para sulfametazina, con valores que exceden los LRM permitidos por el Codex Alimentario.es_ES
