Descripción : The application of chemotherapy based on the use of drugs from the fluoropyrimidine family (5-fluorouracil and capecitabine) is widely used in the treatment of malignant gastro-intestinal tumors, prostate, cervix, breast, among others in Ecuador. The level of response to the use of these drugs in chemotherapeutic treatments is not the same at the population level and may be due to unaware of the pharmacological response that is associated with variation within populations. These variations are associated with the reduction of the enzymatic activity of dipyrimidine dehydrogenase (DPD), the main metabolizer of the fluoropyrimidine family of drugs. The identification of genetic variants of the DPYD gene that codes for the DPD enzyme can provide a basis for customizing the chemotherapeutic tables according to the patient. In this study, 418 individuals including mestizos, Afro-Ecuadorians and indigenous populations were analyzed. The 4 most prevalent variants of the DPYD gene worldwide were genotyped by Sanger sequencing: DPYD 2A (rs3918290), 2846A T (rs67376798), 1679T G (rs55886062) and 1236G A (rs56038477). Allelic frequencies were calculated and correlated with the proportion of ancestry that allowed identifying which variants of the DPYD gene can be associated with different populations worldwide. Finally, the data obtained demonstrate that certain variants of the DPYD gene may be more present in Native American or Afro-descendant populations.
