dc.descriptionThe andean blackberry (Rubus glaucus B.), native to the Andes, is considered one of the most commercialized red fruits in South America for its delicious sweet taste. In Ecuador it is found in wild or cultivated form, and is consumed in jellies, juices or simply as fruit. Interest in this berry, like other red fruits, has increased because of its bioactive compounds that have been linked to potential health benefits due to their anti-inflammatory and antioxidant properties. In this project we investigated the anti-inflammatory properties of blackberry in RAW 264.7 (mouse macrophages) cells by analyzing the relative gene expression of several inflammation and oxidative stress associated genes. To do so, murine macrophages were used, capable to demonstrate a pro-inflammatory phenotype in a process called M1 polarization stimulated by TLRs ligands. The macrophages underwent 5 different treatments in which they were stimulated with a pro-inflammatory agent known as LPS, which acts as a TLR4 ligand, activating signaling pathways that trigger the production of pro-inflammatory cytokines and chemokines; in addition to the anti-inflammatory agent (blackberry extract). Aditionally, RT-qPCR was performed for relative gene expression analysis, using primers for 8 different genes of interest (TNF-α, IL-1b, NFkb, OGG1, iNOS, SOD1, SOD2m, CAT). Our results revealed that the andean blackberry has a preventive, anti-inflammatory activity significantly decreasing NF-κB expression levels, as well as exhibiting an anti-inflamatory tendency in the expression of other genes in cells initially stimulated for 24 hours with blackberry extract followed by 24 hours of LPS treatment.en
