dc.descriptionAntioxidants are beneficial compounds for health by preventing the negative effects of reactive oxygen species (EROs) on normal physiological functions. They are naturally found in fruits, vegetables and everyday foods. Recently, an interest has emerged in the search for antioxidants in other sources of natural origin. That is why, in recent years, microalgae have captured attention as they are able to synthesize bioactive compounds thanks to the biochemical and molecular strategies they have developed to survive in stressful conditions. Due to the lack of knowledge about microalgae in Ecuador, biodiversity and biotechnological applications that confer this group of microorganisms for science have been investigated. From the microalgal isolates of several Protected and Preserved Areas of Ecuador, an evaluation of 129 microalgae isolates from the UDLA Collection of Ficology was carried out in this investigation, from which 19 different strains or Operational Taxonomic Units (OTUs) were selected from molecular identification. The chemical profile was evaluated from methanolic extracts through the study of the total content of phenols, flavonoids, tannins, anthocyanins and chlorophyll a. Similarly, the uptake capacity of the free radical 2,2-diphenyl-1-picrylhydrazyl (DPPH) and the reducing power was evaluated by means of the FRAP (Ferric Reducing Antioxidant Power) test. For the determination of the content of vitamin C and beta-carotene and lutein carotenoids, the High Efficiency Liquid Chromatography (HPLC) method was used. The results of the correlation between the total phenolic content and the antioxidant capacity attribute to these compounds the antioxidant activity of the microalgae. Of the 19 isolates, Chlorella vulgaris had the highest concentration of flavonoids; Acutodesmus obliquus of tannins; Parachlorella kessleri stands out for its anthocyanin content and the strain that presented the highest chlorophyll concentration was Stichococcus sp. From the spectrophotometric tests, it was determined that Scenedesmaceae sp. It is a promising strain because it contains the highest polyphenolic content and reducing capacity, so it is suggested to complement HPLC assays. In general, all strains analyzed have the same or higher concentration of carotenoids compared to fruits, with Scenedesmus vacuolatus and Chlorella vulgaris standing out. Finally, it was determined that Chlorella sorokiniana contains the highest concentration of vitamin C.en
dc.description.abstractLos antioxidantes son compuestos beneficiosos para la salud al prevenir los efectos negativos de Especies Reactivas de Oxígeno (EROs) sobre las funciones fisiológicas normales. De manera natural se encuentran en frutas, verduras y alimentos de consumo cotidiano. Recientemente, ha surgido un interés en la búsqueda de antioxidantes en otras fuentes de origen natural. Es por esto que, en los últimos años las microalgas han captado la atención ya que son capaces de sintetizar compuestos bioactivos gracias a las estrategias bioquímicas y moleculares que han desarrollado para sobrevivir en condiciones de estrés. Debido al escaso conocimiento sobre microalgas en el Ecuador, se ha venido investigando la biodiversidad y las aplicaciones biotecnológicas que confieren este grupo de microorganismos para la ciencia. A partir de aislados microalgales de varias Áreas Protegidas y Conservadas del Ecuador, se realizó en la presente investigación una evaluación de 129 aislados de microalgas de la Colección de Ficología-UDLA, de las que se seleccionaron 19 cepas o Unidades Taxonómicas Operacionales (OTUs) diferentes a partir de la identificación molecular. Se evaluó el perfil químico a partir de extractos metanólicos a través del estudio del contenido total de fenoles, flavonoides, taninos, antocianinas y clorofila a. Del mismo modo, se evaluó la capacidad de captación del radical libre 2,2-difenil-1-picrilhidrazilo (DPPH) y el poder reductor mediante el ensayo FRAP (Ferric Reducing Antioxidant Power). Para la determinación del contenido de vitamina C y los carotenoides beta-caroteno y luteína se empleó el método de Cromatografía Líquida de Alta Eficacia (HPLC). Los resultados de la correlación entre el contenido fenólico total y la capacidad antioxidante atribuyen a estos compuestos la actividad antioxidante de las microalgas. De los 19 aislados, Chlorella vulgaris presentó la mayor concentración de flavonoides; Acutodesmus obliquus de taninos; Parachlorella kessleri destaca por su contenido de antocianinas y la cepa que presentó mayor concentración de clorofila a fue Stichococcus sp. A partir de los ensayos espectrofotométricos, se determinó que Scenedesmaceae sp. es una cepa prometedora al contener el mayor contenido polifenólico y capacidad reductora por lo que se sugiere complementar los ensayos de HPLC. De manera general, todas las cepas analizadas presentan igual o mayor concentración de carotenoides en comparación a frutos, destacándose Scenedesmus vacuolatus y Chlorella vulgaris. Finalmente, se determinó que Chlorella sorokiniana contiene la mayor concentración de vitamina C.es_ES
