dc.descriptionThe protozoa parasite of the genus Leishmania produce high impact diseases named leishmaniases. Mainly, this genus contains 2 subgenus (Viannia and Leishmania) that have 21 pathogenic species for humans. In Ecuador, it has been detected the 2-subgenus presence with the species: L. mexicana, L. amazonensis and L. major (Leishmania subgenus); L. braziliensis, L. panamensis, L. guyanensis, L. naiffi and L. lainsoni (Viannia subgenus). There is a species predominance according to the geographical regions located in the country. It is known that many subgenus-species are closely related with the clinical manifestations in the affected patients as well as the disease’s forecast and the respective downstream treatment. For this disease diagnosis, the most used technique is light microscopy that identifies the Leishmania amastigotes forms. The microscopic technique has a huge disadvantage that is the inability for the differentiation of the subgenus and species, due to, there is no a morphological difference between them. In the case of the detection between subgenus and species in Leishmania, there are some biochemical, immunological and molecular characteristics that are mainly considered. Some molecular tools have been demonstrated to be useful for the diagnosis, characterization, typing and quantification of the etiological agents. In the present investigation, we designed a multiplex-PCR with the main objective to differentiate between the Leishmania and Viannia subgenus. To achieve this objective, we made a bioinformatics analysis that encompassed multiple sequences alignments from different molecular markers with the purpose of obtaining some consensus sequences for each subgenus. In base of this pre-analysis we determined that the cpb and nagA genes are optimum for the differentiation, coinciding with different studies asseverations. With the selection of gen, we designed some discriminant primers that allowed us to develop a downstream multiplex-PCR. As a result, we obtained some PCR products that have the following sizes; one 300 bp amplicon from the nagA gene for the Leishmania subgenus and a 172 pb amplicon from the cpb gene for the Viannia subgenus. The PCR results were confirmed by a later sequencing analysis that demonstrated a high reliability in the developed technique. The standardized technique was applied in immobilized human samples in FTA papers from the Ecuadorian amazon. The present investigation will allow the precise subgenus identification in different patient lesions, as well as the discrimination of reservoirs and vectors. In the future, this technique could help in the development of some projects that are directed to the study of the subgenus geographical distribution.en
