dc.descriptionTemporary immersion systems (SITs) have been designed and automated as a useful tool to provide optimal development conditions that supplies adequate environments for the development of the explant. Some benefits over the use of SIT compared to conventional in vitro propagation are: best provision of nutrients, better gases transfer, a physiological disorders reduction, preservation of morphological explants integrity, among others. There was used a mechanism that used scheduled intervals for immersion in liquid medium, that then goes to a drying phase, where these will stay most of the time. Throughout history, many SITs have been developed, for which different crops have been used in order to test their optimal functioning and also to favor in vitro micropropagation of the different plant species. In the present study, Nicotiana tabacum was used as an experimental model, as it is a model plant in scientific research. Which is currently cultivated around the world because of it high commercial value, mainly as tobacco component. Main objectives of the present study were the design of the temporary immersion system with the capacitance of regulate the immersion time according to the requirements of the plant and the validation of in vitro culture conditions from Nicotiana tabacum buds in the designed immersion system. For which, axillary buds of N. tabacum of a sterile plant material were used. The SIT was programmed for two immersion times (5 min and 15 min) and two immersion frequencies (3 h and 8 h). The studied response variables were: leaves number, shoot length and multiplication coefficient. The experiment was run during a period of 7 days, in which the correct functioning of the SIT was verified. In addition, there were no explants contamination, hyperhydricity, or oxidation. It was determined that the best treatment of the experiment was 6 with 15 g of sucrose, 0.5 x MS, 40 mL / explant, 28 degrees Celsius and an immersion time of 5 min every 8 h. However, for some response variables there were no greater difference between treatment 6 and the others.en
