Resumen : La tipificación de STRs en el cromosoma X se ha convertido en una herramienta extremadamente útil, complementaria a la tipificación de STRs autosómicos, para resolver casos complejos de interés forense en filiación y criminalística…
Descripción : X chromosome STR typing has become a very useful tool in solving complex forensic casework, complementary to autosomal STRs typing. In this study, we genotyped 12 X-STR markers (included in Investigator Argus X-12 and Investigator Argus X-12 QS kits, Qiagen Co.) in 195 women and 167 men, who were part of 147 trios (mother-father-daughter) from Ecuadorian admixed population in order to determine genetic-population, forensic statistics, analyze incompatibilities and de novo mutations. No Hardy-Weinberg equilibrium deviations were observed, except for DXS10135 marker. A significant association was found comparing linkage disequilibrium for all male population loci pairs, after applying the Bonferroni correction, except for DXS10103 – DXS10101, DXS10101 – HPRTB y DXS7423 – DXS10146 pairs. Allele frequency as well as forensic parameters for each X-STR were calculated. There are no significant differences in the X-STR allelic frequencies between women and men. Furthermore, the probability of exclusion, content of polymorphic information and power of discrimination (women and men), combining values from these 12 X chromosome markers were highly informative. Finally, the data showed a range of de novo loci mutation between 2.60 and 7.90 x10-3. Due to the results obtained in this study, we can sustain that the Investigator Argus X-12 and X-12 / QS kits are applicable to the Ecuadorian population in forensic routine analysis cases.
