Descripción : | In 1983, Harris and Mason described a tilting machines system, that they called auxophyton. From the work of these authors, a wide range of semi-automated systems have been developed, which comply with a constant immersion, ideal oxygen transfer, frequent culture medium changes, contamination risk reduction and production costs minimization, which are called temporary immersion systems (TIS), generated in response to the problems of in vitro culture conventional methods. However, despite the fact that the TISs developed to date have allowed to solve several of these problems, each one has disadvantages that make it necessary to develop a TIS that allows to improve results, in order to obtain quality plants. Thus, in this study we analyze and compare Nicotiana tabacum axillary multiplication, an in vitro culture plant species model, in a new temporary immersion system, compared to semi-solid medium culture. Treatments include a temporary immersion system with MS basal medium, another with MS basal medium supplied with 6-Benzylaminopurine (6-BAP, 2 mg - L) and 1-Naphthaleneacetic acid (NAA, 0.2 mg - L) and semisolid systems, which include basal MS medium and another, with MS basal medium supplied with 6-BAP (2 mg - L) and NAA (0.2mg - L). The results showed that there are significant differences between the results obtained in the TIS versus the semisolid medium, both for the basal media without phytohormones, and for the basal media with phytohormones. Demonstrating that the system developed allows obtaining better results that in semi-solid medium, both in the shoots number (14 - 1,05 shoots), leaves number (10 - 0,82 leaves), leaves length (1,94 - 0,23 cm) and multiplication rate (0.81 - 0.06 shoots - day). Therefore, this study represents a starting point for the evaluation and optimization of this system in the future. |
